INGELHEIM, Germany - Monday, June 2nd 2014 [ME NewsWire]
Non-small cell lung cancer (NSCLC) patients with the del19 EGFR mutation lived a median of more than one year longer if they started treatment with afatinib rather than standard chemotherapy1
Afatinib is the first treatment to demonstrate an overall survival benefit for patients with specific types of EGFR mutation-positive NSCLC1
2014 ASCO Annual Meeting
(BUSINESS WIRE)-- For Ex-US and Ex-UK Media only
Boehringer Ingelheim today announced results of the pre-specified individual, as well as the exploratory combined, analyses of two Phase III trials (LUX-Lung 3 and LUX-Lung 6). These data, to be presented at the 50th Annual Meeting of the American Society of Clinical Oncology (ASCO), demonstrated for the first time that patients with NSCLC with the most common epidermal growth factor receptor (EGFR) mutation (exon 19 deletions; del19) lived more than one year longer if treated with first-line afatinib compared to chemotherapy.1
Del19 is the most common type of EGFR mutation. It accounts for 50% of all EGFR mutations. The prevalence of EGFR mutations is approximately 40% among Asian patients and 10-15% among Caucasian patients with lung adenocarcinoma.2,3,4 For patients with the del19 mutation afatinib prolonged survival by 12.2 months versus chemotherapy in the LUX-Lung 3 trial, and 13.0 months versus chemotherapy in the LUX-Lung 6 trial. 1
Principal investigator Professor James Chih-Hsin Yang, M.D., Ph.D., National Taiwan University Hospital in Taiwan commented: “NSCLC patients with the del19 EGFR mutation lived a median of more than one year longer if they started treatment with afatinib rather than standard chemotherapy. Afatinib is the first treatment to demonstrate an overall survival benefit for these patients. LUX-Lung 3 and LUX-Lung 6 represent a potential change in the use of personalised medicine for treating patients with NSCLC. I think that these data support afatinib as the treatment of choice for patients with the del19 mutation.”
Afatinib: approved and investigational indications
Afatinib (GIOTRIF® / GILOTRIF®) is indicated for the treatment of distinct types of EGFR mutation-positive NSCLC. Afatinib is approved in a number of markets, including the EU, Japan, Taiwan and Canada under the brand name GIOTRIF® and in the U.S. under the brand name GILOTRIF®. It is under regulatory review in other countries. Phase III trials in squamous head and neck cancer (HNSCC) and trials in other tumour types are ongoing.

Hilton Worldwide Introduces New Brand: Curio - A Collection by Hilton
Global Collection to Include Highly Anticipated SLS Las Vegas Hotel & Casino
MCLEAN, Va. - Monday, June 2nd 2014 [ME NewsWire]
(BUSINESS WIRE) - Hilton Worldwide today announced the launch of Curio – A Collection by Hilton, including plans for the first hotels to join the new brand.
Created for travelers who seek local discovery and authentic experiences, Curio will be a carefully selected, global collection of distinctive four to five star hotels. Just as the word “curio” can refer to something of interest, unique or even rare, each Curio hotel will be different from the next, with individuality being a common thread, along with the quiet reassurance of the Hilton name behind every location. Letters of intent have been signed for the following properties: SLS Las Vegas Hotel & Casino; The Sam Houston Hotel in Houston, Texas; Hotel Alex Johnson in Rapid City, S.D.; The Franklin Hotel in Chapel Hill, N.C.; and a soon to be named hotel development in downtown Portland, Ore.
“Our customers and owners have expressed strong interest in a brand from us that includes hotels that can retain their unique identity but also deliver the many benefits of our system, including our Hilton HHonors guest loyalty program,” said Christopher J. Nassetta, president and CEO of Hilton Worldwide. “That desire, combined with the current size of the independent hotel market and demand for differentiated hotel stays around the world, made for the right timing for this launch. Curio is an exciting new brand for us – one of two unique concepts we’ll introduce this year.”
Jim Holthouser, executive vice president, global brands, Hilton Worldwide, said, “Curio expands our portfolio to a total of eleven exceptional brands and creates additional opportunities to serve our existing customers even better, attract new customers and grow our pipeline. With Curio, we can help owners leverage our scale, scope and expertise while keeping intact each hotel’s individual character.”
SLS Las Vegas, owned and operated by SBE, is located on the northern end of the Las Vegas Strip at the site of the former Sahara Hotel & Resort – a location steeped in Las Vegas history. The hotel and casino will open following a three-year, $415 million renovation, which includes the total redevelopment of three existing Sahara towers. Billed as a high-style collaboration between SBE Chief Executive Officer Sam Nazarian, renowned design icon Philippe Starck and James Beard Foundation award-winning chef José Andrés, the re-imagination of this 2.5 million square foot development will feature an all-encompassing, mixed-use resort and casino with more than 1,620 guest rooms and suites, 30,000 square feet of flexible meeting space and a collection of SBE’s acclaimed dining and nightlife brands when it opens later this year in time for the September U.S. Labor Day holiday weekend.
Nazarian said of SLS Las Vegas being one of the first Curio hotels, “Today’s announcement marks the beginning of an important relationship between SBE and Hilton Worldwide and an opportunity for me to work alongside a mentor and good friend in Chris Nassetta. Our mission to redefine excellence in hospitality to our guests at SLS Las Vegas aligns perfectly with the Hilton Worldwide vision for Curio. We have a proven record of creating brands that provide memorable guest experiences, whether in the hospitality, dining, nightlife or real estate sectors. Partnering with Hilton and Curio allows us to maintain the SLS brand identity while offering the benefits of Hilton Worldwide’s commercial engines and the strength of the Hilton HHonors loyalty program.”
Additional Details for Planned Curio Hotels
Originally opened in 1924, The Sam Houston Hotel is listed on the National Register of Historic Places. For 90 years, “The Sam” has treated guests to an authentic Texas hospitality experience in downtown Houston. American Liberty Hospitality will maintain management of the hotel when it joins the brand later this year.
Located near the Mount Rushmore National Memorial, Hotel Alex Johnson is also on the National Register of Historic Places and has been a well-known fixture in downtown Rapid City since 1928. Prior to joining Curio next year, the hotel will update its guest rooms, public areas and meeting rooms as part of a multi-million dollar renovation. The Alex Johnson is owned by Cortez LLC, and operated by Isis Hospitality LLC.
Owned and operated by Wintergreen Hospitality, The Franklin Hotel is located in downtown Chapel Hill, one block from the University of North Carolina campus. It is expected to join the brand in late 2014.
Construction is planned to begin in early 2015 on a hotel development by The Widewaters Group Inc., in downtown Portland, Ore. The hotel is anticipated to debut as part of the collection in late 2016.
As members of the Hilton Worldwide portfolio, Curio hotels will offer guests the benefits of Hilton HHonors, the award-winning guest-loyalty program serving more than 40 million members. Hilton HHonors offers more ways to redeem points than any other guest-loyalty program.

Post-Hoc Analysis of Overall Survival in Phase III MPACT Study of Patients with Advanced Pancreatic Cancer Presented at ASCO 2014
ME NewsWire/Business Wire
SUMMIT, N.J. - Monday, June 2nd 2014
2014 ASCO Annual Meeting
Celgene Corporation (NASDAQ: CELG) today announced updated Overall Survival (OS) results from a post-hoc analysis of its phase III MPACT (Metastatic Pancreatic Adenocarcinoma Clinical Trial) study of ABRAXANE® (paclitaxel protein-bound particles for injectable suspension) (albumin-bound) in combination with gemcitabine in treatment-naïve patients with metastatic pancreatic cancer. A poster discussion of the analysis is scheduled for Sunday, June 1st at 11:30 am CT at the 50th American Society of Clinical Oncology (ASCO) annual meeting in Chicago, Ill.
The extended data cutoff occurred at final database lock in May 2013 and allowed for an OS analysis of mature data from 90% of the patients in the study. The extended analysis showed that ABRAXANE plus gemcitabine demonstrated an improvement in OS in the intent-to-treat population compared to patients that received gemcitabine alone [(median OS of 8.7 vs. 6.6. months) (HR0.72, P<0.0001), a difference of 2.1 months]. The analysis showed survival up to 3.5 years in the ABRAXANE plus gemcitabine group (3% of patients alive vs 0% with gemcitabine alone). One- and 2- year survival rates were consistent with the primary analysis.
The analysis also showed that the treatment effect on OS for pre-specified subgroups analyzed in the trial remained consistent across patient subgroups. Specifically, patients with Karnofsky Performance Status (KPS) KPS 90-100 had a higher median OS on ABRAXANE plus gemcitabine as compared to gemcitabine alone [median OS 9.7 months vs. 7.9 months (HR 0.77, P=0.0053)]. Patients with KPS 70-80 also maintained a benefit [median OS 7.6 months vs. 4.3 months (HR 0.59, P<0.0001)].
This updated analysis also evaluated the prognostic effects of CA19-9 and neutrophil-to-lymphocyte ratio (NLR). Both elevated CA19-9 and elevated NLR were associated with poorer survival. Further, treatment with ABRAXANE plus gemcitabine appeared to reduce the effect of CA19-9 as a poor prognostic factor, as similar overall survival was observed regardless of CA19-9 level.
In the MPACT study, the most common grade = 3 treatment-related adverse events in the study for ABRAXANE plus gemcitabine vs. gemcitabine alone were neutropenia, peripheral neuropathy and fatigue. In the ABRAXANE plus gemcitabine arm 17% of patients had grade 3 peripheral neuropathy (no cases of grade 4; 54% had any-grade peripheral neuropathy). Seven percent of patients on the ABRAXANE plus gemcitabine arm that received the average treatment duration experienced Grade 3 neuropathy. The median time to improvement of grade 3 peripheral neuropathy to grade = 1 was 29 days, and 44% of patients resumed treatment with ABRAXANE. Grade = 3 fatigue occurred in 18% of patients. The primary results of the MPACT study from the pre-specified analysis from September 2012 were published in the New England Journal of Medicine in the October 31st 2013 edition.
Details of the poster presentation at ASCO:
Abstract: #4027/Poster #46: Analyses of updated overall survival (OS) and prognostic effect of neutrophil-to-lymphocyte ratio (NLR) and CA 19-9 from the phase III MPACT study of nab-paclitaxel (nab-P) plus gemcitabine (Gem) versus Gem for patients (pts) with metastatic pancreatic cancer (PC). Poster: Sunday, June 1st, 8:00–11:00AM CDT, Location: E 354b; Poster discussion: Sunday, June 1st, 11:30AM–12:45PM CDT, Location: E Hall D2.
These results are from a post-hoc investigational analysis.
About the MPACT Study
In the MPACT (Metastatic Pancreatic Adenocarcinoma Clinical Trial) study, a Celgene-sponsored, open-label, randomized, international study, a total of 861 patients were randomized 1:1 (431 patients to the ABRAXANE/gemcitabine group and 430 patients to the gemcitabine group). The primary endpoint for the study was overall survival. Secondary endpoints were progression-free survival and overall response rate determined by independent radiological review. Other endpoints included progression-free survival and overall response rate as determined by the investigator, and the safety and tolerability of the combination in this patient population.
About Pancreatic Cancer
Pancreatic cancer is the fourth-leading cause of cancer-related death in the U.S. and Europe. The pancreas is composed of two main cell types: exocrine and endocrine. Adenocarcinoma is a sub-type of exocrine tumors and accounts for about 95% of cancers of the pancreas. For all stages of pancreatic cancer combined, the five-year survival rate in U.S. is about 6% and 5.7% in the EU. For metastatic pancreatic cancer, the five-year survival is approximately 1% in the U.S.
About ABRAXANE®
ABRAXANE is an albumin-bound form of paclitaxel that is manufactured using patented nab® technology. ABRAXANE is formulated with albumin, a human protein, and is free of solvents.
In September 2013, the U.S. FDA approved ABRAXANE as first-line treatment of patients with metastatic adenocarcinoma of the pancreas, in combination with gemcitabine. In December 2013, ABRAXANE in combination with gemcitabine was approved for first-line treatment of adult patients with metastatic adenocarcinoma of the pancreas in Europe.
Important Safety Information Based on Approved U.S. Label
WARNING - NEUTROPENIA
?
Do not administer ABRAXANE therapy to patients who have baseline neutrophil counts of less than 1500 cells/mm3. In order to monitor the occurrence of bone marrow suppression, primarily neutropenia, which may be severe and result in infection, it is recommended that frequent peripheral blood cell counts be performed on all patients receiving ABRAXANE
?
Note: An albumin form of paclitaxel may substantially affect a drug’s functional properties relative to those of drug in solution. DO NOT SUBSTITUTE FOR OR WITH OTHER PACLITAXEL FORMULATIONS
CONTRAINDICATIONS
Neutrophil Counts
ABRAXANE should not be used in patients who have baseline neutrophil counts of < 1500 cells/mm3
Hypersensitivity
Patients who experience a severe hypersensitivity reaction to ABRAXANE should not be rechallenged with the drug
WARNINGS AND PRECAUTIONS
Hematologic Effects
Bone marrow suppression (primarily neutropenia) is dose-dependent and a dose-limiting toxicity of ABRAXANE. In a clinical study, Grade 3-4 neutropenia occurred in 38% of patients with pancreatic cancer
Monitor for myelotoxicity by performing complete blood cell counts frequently, including prior to dosing on Days 1, 8, and 15 for pancreatic cancer
Do not administer ABRAXANE to patients with baseline absolute neutrophil counts (ANC) of less than 1500 cells/mm3
In patients with adenocarcinoma of the pancreas, withhold ABRAXANE and gemcitabine if the ANC is less than 500 cells/mm3 or platelets are less than 50,000 cells/mm3 and delay initiation of the next cycle if the ANC is less than 1500 cells/mm3 or platelet count is less than 100,000 cells/mm3 on Day 1 of the cycle. Resume treatment with appropriate dose reduction if recommended
Nervous System
Sensory neuropathy is dose- and schedule-dependent
The occurrence of Grade 1 or 2 sensory neuropathy does not generally require dose modification
If = Grade 3 sensory neuropathy develops, withhold ABRAXANE treatment until resolution to = Grade 1 followed by a dose reduction for all subsequent courses of ABRAXANE
Sepsis
Sepsis occurred in 5% of patients with or without neutropenia who received ABRAXANE in combination with gemcitabine
Biliary obstruction or presence of biliary stent were risk factors for severe or fatal sepsis
If a patient becomes febrile (regardless of ANC), initiate treatment with broad-spectrum antibiotics
For febrile neutropenia, interrupt ABRAXANE and gemcitabine until fever resolves and ANC =1500 cells/mm3, then resume treatment at reduced dose levels
Pneumonitis
Pneumonitis, including some cases that were fatal, occurred in 4% of patients receiving ABRAXANE in combination with gemcitabine
Monitor patients for signs and symptoms and interrupt ABRAXANE and gemcitabine during evaluation of suspected pneumonitis
Permanently discontinue treatment with ABRAXANE and gemcitabine upon making a diagnosis of pneumonitis
Hypersensitivity
Severe and sometimes fatal hypersensitivity reactions, including anaphylactic reactions, have been reported
Patients who experience a severe hypersensitivity reaction to ABRAXANE should not be rechallenged with this drug
Hepatic Impairment
Because the exposure and toxicity of paclitaxel can be increased with hepatic impairment, administration of ABRAXANE in patients with hepatic impairment should be performed with caution
For pancreatic adenocarcinoma, ABRAXANE is not recommended for patients with moderate or severe hepatic impairment
Albumin (Human)
ABRAXANE contains albumin (human), a derivative of human blood
Use in Pregnancy: Pregnancy Category D
ABRAXANE can cause fetal harm when administered to a pregnant woman
If this drug is used during pregnancy, or if the patient becomes pregnant while receiving this drug, the patient should be apprised of the potential hazard to the fetus

Women of childbearing potential should be advised to avoid becoming pregnant while receiving ABRAXANE
Use in Men
Men should be advised not to father a child while receiving ABRAXANE

(BUSINESS WIRE)-- David Sable, Global CEO of Y&R, announced today that he’s promoting Michael Sussman to President of BAV® Worldwide, Y&R. Sussman has been with Y&R for 17 years and was most recently the Global Director of Analytic Insights and Strategy for Y&R Advertising. Michael oversees BrandAsset® Valuator, Y&R’s proprietary brand model and global database, along with other quantitative tools essential to the agency’s strategic process. He will now lead the charge of integration and growth of BAV® throughout the agency.
“Michael has been part of the development of BAV® since shortly after its inception,” said Sable. “He is deeply committed to the agency, full of passion for brands and masterful at interpreting data to tell compelling stories about them.
Michael has been part of our BAV® team since its early days and is absolutely the best person to help us drive BAV® forward in key markets. This is a shining moment for BAV®, now in its 20th year; it remains the largest database with consumer perceptions of brands, and Michael will be fundamental in continually sustaining and differentiating its relevance in the market.”
In addition to working with Sandy Thompson, Y&R’s Global Planning Director, to continue to drive integration of BAV® and analytics into Y&R’s strategic process, consulting across agency clients and driving thought leadership, Sussman will also oversee the global BAV® research and fielding process. Sussman has been tasked with increasing the amount of “brandscape” studies being conducted in important markets; with eight new market studies already in progress this year.
Michael has also organized a team of regional strategic leads in key Y&R markets (“hubs of expertise”) to help guide the global fielding process. This team will foster greater collaboration and sharing of thought leadership across the Y&R network, as well as helping to drive deeper BAV® consulting relationships with Y&R clients around the world.
“It’s an honor to take the next step in my Y&R career, and I couldn’t be more excited about the future of BAV® as we work with strategic leads across the network to raise the impact of our studies and data driven insights to new heights,” said Sussman. “I’m thrilled to take my 17 years of experience designing research, interpreting data, and exploring brand equity for Y&R’s clients with BAV®, and integrating these learnings on a global level.”
Sussman and his team will continue to partner with John Gerzema, Chairman and CEO of BAV® Consulting and his team to build greater synergies between Y&R Advertising and BAV® Consulting, which has its own client base.
"Michael and I go back a long way,” said Gerzema. “He has lived and breathed BAV for so many years, which helps him to continually bring new understanding of the data and the BAV model. We are looking forward to Michael’s development of the regional hubs across Y&R. I know that our clients will benefit from this new infrastructure and really look forward to collaborating with him again in his new role."

NEW YORK - Saturday, May 31st 2014 [ME NewsWire]
