Merck Highlights New Four-Year Efficacy and Safety Data for Investigational BTK Inhibitor, Evobrutinib, in RMS

 

  • Data from the ongoing Phase II extension showed treatment benefits of evobrutinib were maintained over four years and remained consistent with the efficacy and safety profile seen in earlier data

Merck, a leading science and technology company, today announced updated long-term efficacy and safety data for investigational evobrutinib that continue to show a favorable safety and tolerability profile, consistent with what was seen earlier in the double-blind period (DBP) of the clinical trials. These data also continue to demonstrate its treatment benefit in reducing annualized relapse rates (ARR) over four years in people with relapsing multiple sclerosis (RMS). Data will be presented at the Americas Committee for Treatment and Research in Multiple Sclerosis (ACTRIMS) Forum 2023, taking place February 23-25.

Data from the ongoing Phase II open-label extension (OLE) trial of evobrutinib showed that treatment benefits were maintained over four years, with no new safety signals. Pooled ARR at week 228 of the OLE for all patients across the original DBP dosing groups was 0.13, with a further reduction seen in the period after switching from evobrutinib 75mg once-daily to 75mg twice-daily, from 0.19 to 0.10. For those receiving 75mg twice-daily dosing in the DBP, ARR was 0.11 at the end of DBP and 0.12 at week 228 of OLE. These data further support the twice-daily dosing currently being examined in Phase III clinical trials.

Overall, treatment emergent adverse events (TEAEs) were mild/moderate in the OLE, with 3.3% (n=7) of patients with RMS experiencing a serious TEAE. No new safety signals were seen in the OLE and evobrutinib continued to show consistent tolerability after up to four years of treatment. There was no dose dependent increase in TEAEs observed in patients who switched to twice-daily evobrutinib 75mg in the OLE.

Additionally, at ACTRIMS, data will be presented that includes analyses of the CLARIFY-MS study, showing the potential of MAVENCLAD® (cladribine tablets) to improve outcomes in an impactful way for people living with RMS.

  • Participants maintained their employment status over the two years of the study, which included the COVID-19 pandemic, with 43.4% (n=209) of patients employed full-time at month 24, compared to 47.5% (n=229) at baseline.
  • Cognitive function remained stable over two years of treatment as measured at baseline, 12 and 24 months through the Brief International Cognitive Assessment for MS (BICAMS) battery, which is comprised of tests of mental processing speed and memory.

About Evobrutinib

Evobrutinib is an oral, CNS-penetrating, highly selective inhibitor of Bruton’s tyrosine kinase (BTK) in clinical development as a potential treatment for relapsing multiple sclerosis (RMS). It is the first BTK inhibitor to demonstrate clinical efficacy in the largest Phase II study with follow-up beyond three years as well as demonstrate an impact on early biomarkers of ongoing central inflammation that correlate with disease progression, including slowly expanding lesions (SEL) volume and levels of blood neurofilament light chain protein (NfL). Evobrutinib is designed to modulate B cell responses such as proliferation and antibody and cytokine release, as well as modulate macrophage/microglia activation. During Phase II, the BTKi dose finding study demonstrated that BID dosing achieved maximal efficacy with >95% BTK occupancy maintained in 98% of patients before the next dose. Evobrutinib is currently under clinical investigation and is not approved for any use anywhere in the world.

About the Open-Label Extension (OLE) Phase II Clinical Trial with Evobrutinib

In the 48 week double-blind period (DBP), patients with RMS were assigned to one of five treatment groups: placebo (switching to 25mg once-daily evobrutinib after 24 weeks), 25mg or 75mg once-daily evobrutinib, 75mg twice-daily evobrutinib, or open-label dimethyl fumarate (120mg twice daily for the first week and 240mg twice daily thereafter). At week 48, patients could enter the OLE and received evobrutinib 75mg once daily for a mean time of 49.8 weeks before switching to 75mg twice daily for the remainder of the OLE.

About MAVENCLAD®

MAVENCLAD, approved by the U.S. Food and Drug Administration (FDA) on March 29, 2019, is the first and only short-course oral therapy for the treatment of adults with relapsing-remitting disease (RRMS) and active secondary progressive disease (SPMS). Because of its safety profile, use of MAVENCLAD is generally recommended for patients who have had an inadequate response to, or are unable to tolerate, an alternate drug indicated for the treatment of multiple sclerosis (MS), and MAVENCLAD is not recommended for use in patients with clinically isolated syndrome (CIS). Patients should follow healthcare provider instructions including cancer screening, contraception and blood tests. The approved dose of MAVENCLAD is 3.5 mg per kg body weight over two years, administered as one treatment course of 1.75 mg per kg per year, each consisting of two treatment weeks. The mechanism by which cladribine exerts its therapeutic effects in patients with multiple sclerosis has not been fully elucidated but is thought to involve cytotoxic effects on B and T lymphocytes through impairment of DNA synthesis, resulting in depletion of lymphocytes. MAVENCLAD causes a dose-dependent reduction in lymphocyte counts followed by recovery.

Quectel Announces RedCap Rx255C Module Series to Help Expand the Reach of 5G into More IoT Applications and Verticals

Meeting the 3GPP Release 17 standards, the Rx255C series supports 5G standalone (SA) mode and a maximum bandwidth of 20MHz on the sub-6GHz frequency band. Targeting all markets worldwide, the 5G RedCap modules are backward compatible with LTE networks and cover nearly all the mainstream carriers worldwide. They provide a theoretical peak downlink data rate of around 220 Mbps and uplink data rate of around 100 Mbps, sufficient to meet the demand for data speeds in IoT applications such as robotics, DTU, drones, smart ports, smart grid, AR/VR wearables, educational laptops as well as other entry level mobile broadband devices.


The support of RedCap technology allows the Rx255C to optimize the number of antennas, reduce the transmitting and receiving bandwidth, and provide 64QAM/256QAM (optional) modulation to greatly optimize the cost and size. The high level of integration and unique architecture of Snapdragon X35 ensures that the modules also have a low power consumption which drives 5G adoption in an entirely new category of devices.


The Rx255C series supports L1+L5 dual frequency GNSS to achieve precise positioning services. They provide a wide range of interfaces such as PCIe 2.0, USB 2.0 and supplementary functions including VoLTE and DFOTA.

The first version of XProtect® – Milestone’s open-platform video management software and core product – was sold in 2002 to an end user in Canada. Since then, Milestone’s technology has been deployed in more than 500,000 installations through a network of distributors and resellers. Milestone originated from an idea for seamlessly transferring financial data from the stock exchange. Founders John Blem and Henrik Friborg Jacobsen discovered the same technology could be used to transfer video data.

In addition to selling software, Milestone also provides a hardware product called Husky. The first version was launched in 2013.

(Smart) and Spectrum Effect® announced today that Smart has reached a new standard of network performance with best-in-class network experience for its customers with the deployment of Spectrum-NET.


Smart deployed Spectrum-NET nationwide in 2022, enabling Smart to reduce in half the interference impacting their network, making Smart the cleanest network in the region. According to data from Spectrum Effect, Smart now performs better than other operators in Asia Pacific, the Middle East and Africa, and Latin America, putting Smart’s performance at par with North American and European countries. Smart is presently in the process of turning up Spectrum-NET’s new auto-mitigation capability, immediately increasing network capacity and improving service quality for subscribers affected by remaining interference sources.

Smart launched the Spectrum Interference Extraction Detection (SIED) Program driven by Spectrum-NET as a game-changer in managing and mitigating the impact of RF Interference. The SIED program analyzed Smart's entire cellular network and removed 60% of Passive Intermodulation (PIM) interfered cells and 65% of RAN-Generated Interference (RGI) impacted cells to normal levels as of January 2023. To stem the continued increase in External Interference sources (e.g., substandard repeaters or “boosters”), especially in urban areas, Smart has been working closely with the National Telecommunications Commission (NTC).

Lenovo Launches Annual Intelligent Transformation Index to Explore the Global Adoption of Digital Intelligence


(BUSINESS WIRE) -- To explore the adoption of technology driving intelligent transformation, Lenovo today launched the first edition of the global Intelligent Transformation Index, which benchmarks adoption rates of transformative digital technologies across markets and industries. The report is written by Economist Impact and commissioned by Lenovo.


Across all businesses, technology is transforming every aspect of an organization’s function, but it is technologies such as big data, artificial intelligence (AI), and the internet of things (IoT) that are driving a new paradigm of change – intelligent transformation – for better social and economic outcomes. However, the lack of skilled labor is slowing the speed of adoption.


The Intelligent Transformation Index


Lenovo’s Intelligent Transformation Index ranks market adoption of big data, AI, and IoT, based on published data from the OECD, and proprietary survey data. The team at Economist Impact identified that markets based in Northern and Western Europe showed the highest adoption rates compared to other markets, primarily due to advanced infrastructure, skilled labor force, and forward-looking government policies that promote trust and transparency in data usage.


The highest of all the markets included in the index was Denmark, where the Intelligent Transformation Index showed that 27% of its businesses used big data while 24% had deployed AI technology. While advanced economies generally have made more progress in Intelligent Transformation, a high GDP per capita does not guarantee good performance. Canada and Australia ranked 31st and 32nd, respectively, of the 33 countries studied.


When looking at the adoption of big data, AI, and IoT, across industries, the ICT Services sector shows the highest overall level of adoption. Firms in the manufacturing and logistics industries show high adoption of IoT due to the use case of this specific technology in efficiency-driven supply chain intelligence. Knowledge industries such as professional, scientific, and technical services show more developed use cases for big data and AI.


The least “intelligent” sectors are accommodation, food service, and construction. In these industries, firms tend to be smaller or family-run businesses that lack the scale and resources to deploy the newer wave of technologies.


Where next for Intelligent Transformation?


While digital intelligence is still in its infancy, it is clear from Lenovo’s Intelligent Transformation Index that adoption is wide-scale and cross-industry. While building digital infrastructure can be relatively quick, the biggest barrier to adoption is the availability of a skilled labor force. Equipping employees with the necessary skills to adapt to increasingly data-driven and automated work processes will be a key challenge for organizations in the coming years.


Moving into 2023, companies across the world are focusing on controlling expenses in the face of an uncertain economy. Integrating big data, AI, and IoT as an out-of-the-box solution via the cloud can help support an organization’s goal towards intelligent transformation, helping increase affordability and lowing the barrier to adoption, while driving greater business intelligence.
